Action/Kinetics:
t1/2, absorption: 25-50 hr. t1/2, elimination: 5-20 min. Extensively bound to plasma proteins. Metabolized in liver; excreted through urine and feces.
Uses:
Progesterone supplementation or replacement as part of assisted reproductive technology treatment for infertile women with progesterone deficiency. Secondary amenorrhea.
Contraindications:
Undiagnosed vaginal bleeding, liver disease or dysfunction, known or suspected malignancy of breast or genital organs, missed abortion, active thrombophlebitis or thromboembolic disease (or history of such). Concurrent use with other local intravaginal therapy.
Special Concerns:
Safety and efficacy have not been determined in children.

How Supplied:
Vaginal applicator: 90 mg/1.125 g (Crinone 8%), 45 mg 1.125 g (Crinone 4%)
Dosage
•Vaginal gel: Crinone 8% Assisted reproductive technology.
90 mg once daily for women who require progesterone supplementation. Administer 90 mg b.i.d. in women with partial or complete ovarian failure who require progesterone replacement. If pregnancy occurs, treatment may be continued until placental autonomy has been achieved (up to 10-12 weeks).
•Vaginal gel: Crinone 4%
Secondary amenorrhea.
45 mg every other day up to total of 6 doses. For women who fail to respond, Crinone 8% (90 mg) may be given every other day up to total of 6 doses. |
