]Action/Kinetics:
Intermediate-acting. Is five times more potent than hydrocortisone and cortisone. Minimal side effects except for GI distress. Moderate mineralocorticoid activity. Plasma t1/2: over 200 min.
Contraindications:
Lactation.
Special Concerns:
Use with particular caution in diabetes.
How Supplied:
Prednisolone: Syrup: 5 mg/5 mL, 15 mg/5 mL; Tablet: 5 mg. Prednisolone acetate: Injection: 25 mg/mL, 50 mg/mL; Ophthalmic Suspension: 0.12%, 0.125%, 1%. Prednisolone sodium phosphate: Liquid: 5 mg/5 mL; Ophthalmic Solution: 0.125%, 1%. Prednisolone tebutate: Injection: 20 mg/mL.
Dosage
Prednisolone
•Tablets, Syrup
Most uses.
5-60 mg/day, depending on disease being treated.
Multiple sclerosis (exacerbation).
200 mg/day for 1 week; then, 80 mg on alternate days for 1 month.
Pleurisy of tuberculosis.
0.75 mg/kg/day (then taper) given concurrently with antituberculosis therapy.

•IM
4-60 mg/day. Not for IV use.
Multiple sclerosis (exacerbation).

•Intralesional, Intra-articular, Soft Tissue Injection
4-100 mg (larger doses for large joints).
•Ophthalmic Suspension (0.12%, 0.125%, 1%)
Instill 1-2 gtt into the conjunctival sac b.i.d.-q.i.d. During the first 24 to 48 hr, the frequency of dosing may be increased if necessary.

•IM Only
20-80 mg acetate and 5-20 mg sodium phosphate every few days for 3-4 weeks.
•Intra-articular, Intrasynovial
20-40 mg prednisolone acetate and 5-10 mg prednisolone sodium phosphate.

•PO Solution
Most uses.
5-60 mg/day in single or divided doses.
Adrenocortical insufficiency.
Pediatric: 0.14 mg/kg (4 mg/m2) daily in three to four divided doses.
Other pediatric uses.
0.5-2 mg/kg (15-60 mg/m2) daily in three to four divided doses.
•IM, IV
4-60 mg/day.
Multiple sclerosis (exacerbation).

2-30 mg, depending on site and severity of disease.
•Ophthalmic Solution (0.125%, 1%)
1-2 gtt into the conjunctival sac q hr during the day and q 2 hr during the night; then, after response obtained, decrease dose to 1 gtt/ q 4 hr and then later 1 gtt t.i.d.-q.i.d.
Prednisolone tebutate
•Intra-articular, Intralesional, Soft Tissue Injection
4-30 mg, depending on site and severity of disease. Doses higher than 40 mg are not recommended. |
